      OverviewPlatelet Derived Growth Factor (PDGF) is a potent mitogen for a wide range of cell types including fibroblasts, smooth muscle and connective tissue. PDGF, which is composed of a dimer of two chains termed the A chain and B chain, can be present as AA or BB homodimers or as an AB heterodimer. Human PDGF-AA is a 28.5 kDa A chain homodimer protein containing 250 amino acid residues.

The four members of this family are mitogenic factors for cells of mesenchymal origin and are characterized by a motif of eight cysteines. This gene product can exist either as a homodimer or as a heterodimer with the platelet-derived growth factor beta polypeptide, where the dimers are connected by disulfide bonds. Studies using knockout mice have shown cellular defects in oligodendrocytes, alveolar smooth muscle cells, and Leydig cells in the testis; knockout mice die either as embryos or shortly after birth. Two splice variants have been identified for this gene.

      Storage ConditionsThe lyophilized PDGF-AA is stable for a few weeks at room temperature, but best stored at -80°C. After a quick spin, reconstitute in ddH2O to a concentration of 0.1-1.0 mg/mL. This solution can be diluted into aqueous buffered solutions and stored at 2-8°C for one week or -20°C for future use. Reconstituted PDGF-AA should be stored in undiluted aliquots at -20°C for up to six months.
