A sulfonated azo dye, initially identified by a Trypanosoma burcei RNA-editing ligase 1 (TbREL1) crystal structure-based inhibitor screening, is shown to inhibit TbREL1 activity (IC50 = 1.95 µM) by targeting the enzyme's ATP binding pocket. V1 is less potent against T4 phage RNA ligase 2 and human DNA ligase IIIβ (IC50 = 3.53 and 27.49 µM, respectively).
