      Background InformationInterferon-inducible protein AIM2 (UniProt: O14862; also known as Absent in melanoma 2) is encoded by the AIM2 gene (Gene ID: 9447) in human. The human HIN-200 family proteins consist of four members known as IFI 16, MNDA, AIM2, and IFIX. AIM2 is an IFN- inducible protein that is expressed in spleen, small intestine, peripheral blood leukocytes, and testis. It is involved in innate immune response by recognizing cytosolic double-stranded DNA and induces caspase-1-activating inflammasome formation in macrophages. Its HIN-20 domain is localized in amino acids 138-337, which mediates dsDNA binding via electrostatic interactions. At its N-terminal region it contains a pyrin domain (aa 1-87) and at the C-terminal it has an oligonucleotide/oligosaccharide-binding domain that senses cytoplasmic DNA. It is reported to interact with ASC (apoptosis-associated speck-like protein containing a CARD) through its pyrin domain to activate caspase-1. It detects cytosolic dsDNA of viral and bacterial origin in a non-sequence-specific manner that triggers its oligomerization and formation of AIM2 inflammasome. It also acts as a mediator of pyroptosis, necroptosis, and apoptosis, which are integral parts of host defense against pathogens. Clone 3B10 is shown to specifically recognize AIM2 and does not react with other related member of the HIN-200 family. (Ref.: Vajjhala, PR., et al. (2015). J. Biol.
