      Background InformationMediator of RNA polymerase II transcription subunit 15 (UniProt Q96RN5; also known as Activator-recruited cofactor 105 kDa component, ARC105, CTG repeat protein 7a, Mediator complex subunit 15, PC2 glutamine/Q-rich-associated protein, Positive cofactor 2 glutamine/Q-rich-associated protein, TIG-1, TPA-inducible gene 1 protein, Trinucleotide repeat-containing gene 7 protein) is encoded by the MED15 (also known as ARC105, CTG7A, PCQAP, TIG1, TNRC7) gene (Gene ID 51586) in human. The Mediator complex is an essential transcriptional cofactor of RNA polymerase II (Pol II). The Mediator complex is composed of the CDK/cyclin, the head, the middle, and the tail modules. The tail module consists of eight subunits (MED2/MED29, MED3/MED27, MED5/MED24, MED14, MED15, MED16, MED23, and MED25) and mediates the recruitment of various transcriptional regulators. Human MED15 contains an N-terminal KIX domain (a.a. 9-73) that mediates MED15 binding to the sterol regulatory element binding protein (SREBP) and is important for MED15 function in cholesterol and fatty acid homeostasis. Similar to its yeast counterpart (yeast Gal11), human MED15 is found to colocalize with TFIIE and TFIIF, most likely promoting transcriptional activation in collaboration with these two transcription factors.
