      Background InformationSynembryn-B (UniProt Q80XE1; also known as Protein Ric-8B, Resistance to inhibitors of cholinesterase 8B) is encoded by the Ric8b (also known as BC051080) gene (Gene ID 237422) in murine species. Ric-8A and Ric-8B stimulate nucleotide exchange of all Gα subunit classes by stabilizing the Gα nucleotide-free transition state. Ric-8A acts upon Gαi/q/13, while Ric-8B targets Gαs/olf. In addition to their function as guanine nucleotide exchange factors (GEFs), Ric proteins also act as molecular chaperones that directly participate in the posttranslational folding of nascent Gα subunits to promote G-protein biosynthesis and membrane association. Ric-8 knockdown reduces Gα steady-state abundances and overexpression of Ric-8 proteins enhances cellular Gα levels. In Ric-8A– or Ric-8B–delicient ES cells, newly produced Gα subunits are defective in the initial association with an internal membrane, resulting in a ∼10-fold faster turned over rate than in WT cells.
