The NFKB complex is inhibited by I-kappa-B proteins (NFKBIA, MIM 164008, or NFKBIB, MIM 604495), which inactivate NF-kappa-B by trapping it in the cytoplasm. Phosphorylation of serine residues on the I-kappa-B proteins by kinases (IKBKA, MIM 600664, or IKBKB) marks them for destruction via the ubiquitination pathway, thereby allowing activation of the NF-kappa-B complex.       Quality AssuranceEvaluated by Kinase assay: 10 µl of varying amounts of IKKβ was mixed with 5 µl of 1 mg/ml IKKtide synthetic peptide substrate and 5 µl of distilled water. Reaction was initiated by adding 5 µl of 250 µM [33P]-ATP and incubating for 15 min at 30°C followed by spotting the reaction on phosphocellulose paper, fixing in 1% phosphoric acid, and assaying with a scintillation counter.
