      Background InformationClaudin 5 (UniProt: O00501; also known as Transmembrane protein deleted in VCFS, TMDVCF) is encoded by the CLDN5 (also known as AWAL, TMVCF) gene (Gene ID: 7122) in human. Claudin 5 is a multi-pass membrane protein that plays a major role in tight junction-specific obliteration of the intercellular space. It is localized specifically to the endothelial cell layer in the brain, lung, liver, kidney, and skin. In peripheral tissues, it displays greater heterogeneity in tissue expression. It is one of the most enriched tight junction proteins and its dysfunction has been linked to several neurodegenerative diseases, including Alzheimer s disease and multiple sclerosis. Cldn5-null mice are reported to die shortly after birth. Claudin 5 contains four transmembrane domains (aa 8-28, 82-102, 123-143, 160-180), two extracellular loops (aa 29-81, 144-159), and three cytoplasmic domains (aa 1-7, 103-122, 181-218).
