      Background InformationSeptin 7 (UniProt: F6UVF8; also known as SEPT7, Septin-7) is encoded by the SEPTIN7 gene (Gene ID: 100070492) in horse. Septins are highly conserved, GTP-binding cytoskeletal proteins that are involved in various cellular processes, including cytokinesis, membrane dynamics, cellular morphology, carcinogenesis, and neurodegeneration. Thus far 13 different septins have been reported in mammals. They can self-assemble and form filaments that serve as scaffolds for protein recruitment. Their assembly consists of hexamers and octamers that are formed by septin subunits, all including the essential group member Septin 7. Septin 7 can combine with other septins to form heteropolymers and it is a core component of these multimeric septin complexes. These heteropolymers can form a diverse array of higher order structures, including filaments, gauzes, and rings. Septin 7 is shown to be a novel regulator of neuronal calcium homoeostasis. It is widely distributed in the brain and is reported to be the most common septin in post-synaptic density fractions. It affects cytosolic calcium levels by down regulating the expression of the Orai and IP3 receptors. Lack of septin 7 can lead to loss of other septin proteins in homo- and heterooligomeric complexes. It is also shown to be essential for regulation of dendrite branching and dendritic spine morphology. Reduced levels of septin 7 have been reported in blood-derived lymphocytes in an equine model of recurrent uveitis (ERU) that affects inner eye and leads to vision loss. Dedicator of cytokinesis 8 (DOCK8) protein is shown to be an interaction partner of septin 7 in primary lymphocytes and its levels also show a significant decline in models of ERU.
