      Background InformationThe T cell receptor or TCR is a plasma membrane protein complex responsible for recognizing antigens bound to major histocompatibility complex (MHC) molecules on antigen presenting cells. In 95% of T cells, the TCR is composed of a disulfide-linked alpha (α) and beta (β) dimer, while among 5% of T cells the TCR is composed of a gamma (γ) and a delta (δ) dimer. T cells are activated when their surface TCRs engage with antigen/MHC complex on antigen presenting cells. Each TCR alpha and beta chain contains a variable (V) and a constant (C) extracellular immunoglobulin-like domain, followed by a transmembrane region and a short cytoplasmic tail. The C domain lies proximal to the plasma membrane, while the V domain engages the antigen/MHC complex. Similar to immunoglobulin light chain and heavy chain, the TCR alpha and beta genes (Gene ID 21473 and 21577, respectively, in murine species) undergo VJ and VDJ recombination to generate TCR diversity for recognizing a wide range of antigens. the alpha and beta chains have very short cytoplamic tail and reply on the CD3 (cluster of differentiation 3) co-receptor for transducing signaling further intracellularly.

      Application NotesFlow Cytometry Analysis: A representative lot detected αβ TCR surface expression on 99% of adult lymph node T-cells from mouse strain C3H/HeJ, while only 10% of adult and 1% of newborn thymocyte population were found to be αβ TCR-expressing mature thymocytes (Finkel, T.H., et al. (1989). J. Immunol. 142(9):3006-3012).
      Activity Assay: Representative lots, when immunobilized on well surface, activated murine T cell hybridomas (DO-11.10, 2QK34.7, and A1.1) by cross-linking cell surface αβ TCR. Clone H57-597 did not stimulate T cell hybridomas that express γδ TCR (Shi, Y.F., et al. (1991). J. Immunol. 146(10):3340-3346; (Grégoire, C., et al. (1991). Proc. Natl. Acad. Sci. U. S.
