      OverviewHuman recombinant HDAC6 (His-tag) produced in insect cells.
      Background InformationHistone deacetylases (HDACs) are chromatin-remodeling factors that deacetylate histones and act as transcriptional repressors or silencers of genes. They regulate histone acetylation by catalyzing the removal of acetyl groups on the amino terminal lysine residues of the core nucleosomal histones. Each cell is reported to have several different types of HDACs. HDAC6 is a member of the Class II HDACs that displays limited tissue distribution and can shuttle between nucleus and cytoplasm. In the cytoplasm, it is it is reported to associate with microtubules. HDAC6 contains two leucine-rich nuclear export sequences (NES1, NES2) that play an essential role in the cytoplasmic/nuclear shuttling process. The amino acid residues in HDAC6 are arranged in the space to form two independent catalytic domains with a zinc finger ubiquitin-binding domain located at the C-terminus. HDAC6 is responsible for the deacetylation of lysine residues on the N-terminal part of the core histones (H2A, H2B, H3 and H4).

      Storage and Shipping Information
      Storage ConditionsStore at -80°C. The enzyme is stable on ice for 30 to 60 min, but may lose activity with dilution and/or prolonged storage on ice. It is recommended that thawing and addition of the enzyme to assay wells be done within as short a time as possible before start of the assay and that the thawed enzyme be kept on ice throughout. Best performance will be obtained by adding the chilled, undiluted enzyme directly to pre-warmed buffer and proceeding immediately to the addition of pre-warmed substrate. The remaining, unused enzyme should be refrozen quickly by, for example, snap freezing in a dry/ice ethanol bath or liquid nitrogen. The enzyme is stable to at least 4 freeze/thaw cycles. To minimize the number of freeze/thaw cycles, aliquot the HDAC6 into separate tubes before storing at -80°C.
