Overview
An optimized concentration of various reversible and irreversible inhibitors to inhibit serine, cysteine, and calpain proteases. Suitable for the protection of proteins purified from animal tissues, plant tissues, yeast, and bacteria. EDTA is also provided separately to inhibit metalloproteinases. Protease Arrest™ Reagent is provided as a 100X solution that when diluted in extraction buffer at pH 7.0-8.0 inhibits 95-98% of protease activity. Supplied with a data sheet.

Description
Protease Arrest™ Reagent provides optimized concentration of the various inhibitors, and exhibits excellent inhibitory activity towards various proteases. It is suitable for protection of proteins during purification. It can be used with animal tissues, plant tissues, yeast and bacteria. Protease Arrest™ Reagent contains a blend of both irreversible and reversible inhibitors for serine, and cysteine proteases and for calpains. A vial of EDTA is also provided to inhibit metalloproteases. Please note, however, that some proteins may require divalent cations like Ca2+, Mg2+ or Mn2+ for their activity, as the presence of EDTA may be detrimental to their enzymatic activity. Furthermore, if the protein of interest is purified using immobilized metal chelate affinity chromatography, EDTA must be removed from the buffer before the chromatography. The EDTA solution may be added in the extraction buffer or lysate as needed. Protease Arrest™ Reagent at 1X concentration in extraction buffer at pH 7-8 inhibits over 95% of protease activities. Each set contains the following components

Recommended reaction conditions
1. Allow solution to warm to the room temperature. The solution is in suspension form, vortex the vial before removing the solution.
2. Add Protease Arrest™ Reagent 10 µl/ml directly to an appropriate volume of extraction buffer or extract to 1X final concentration.
3. For greater protease inhibition, add Protease Arrest™ Reagent 20-30 µl/ml to get 2-3X final concentration
4. For inhibition of metalloproteases (if the buffer does not contain EDTA), add EDTA (0.5 M) 10 µl/ml directly in an appropriate volume of extraction buffer or extract to 1X final concentration.
